96-WELL PLATES


MatTek PermaCell 96-well insert plates are advanced, sterile, cell culture devices used for growth and differentiation of cells. The 96-well insert plates consist of an array of 96 membrane wells connected into a single, rigid tray for easier handling and feeding
and to allow for high-throughput/ robotic processing. The 96-well insert plate fits within a 96-well receiver plate (included) which allows for individual treatment and assaying of each well in the plate.

MatTek PermaCell 96-well insert plates can be used for cell culture and/or cell differentiation into 3-dimensional tissues, drug transport/permeability studies, and imaging studies.

Note: When working with MatTek PermaCell insert plates, use aseptic techniques and appropriate culture media, reagents, and growth environments for specific cell types. Depending on the specific insert and cell type, this could include coating the inserts with an extracellular matrix (ECM) or other attachment aids.

NOTE: WPI is an authorized distributor of MatTek for the 96-well cell culture insert plates.

DIRECTIONS FOR USE

To ensure sterility, all steps listed below should be performed in a laminar flow hood or equivalent controlled environment.
1. Inserts come pre-packaged in 96-well receiver plate.
2. Remove the insert plate and inspect the wells to ensure there are no defects.

COATING (OPTIONAL)

1. Coat insert plate with appropriate ECM following manufacturer’s recommendations: for 96-well plate inserts use 25-75 μL/insert.

2. Incubate the plate following manufacturer's recommendations and then remove coating and wash inserts. The insert plates are now ready for use - refer to Plate Set-up below.

Plate Setup

1. Add cell culture media to wells of the 96-well receiver plate: For MatTek 96-well insert plates, add 275-300 μL of medium into the wells of the receiver plate.

2. Seed cells into the wells of the insert plate and follow standard tissue culture incubation and feeding procedures for optimal cell growth.

             a. Typical experiments seed cells on the apical membrane using up to 150μL in the apical compartment.

             b. Experiments can also include seeding cells into the receiver plate wells and/or onto the basal side of the membrane.

Note: For TEER measurements, volumes of 350-400 μL of TEER buffer or PBS in the wells of the receiver plate and 150μL of buffer in the apical compartment are recommended.
